Preparation and characterization of diazepam loaded nanomicelles for pediatric intravenous dose adjustment

The common side effects of lipophilic drug delivery for parenteral is the irritating or burning sensation as results of cosolvent influences on the blood vessels as well the precipitation on dilution with intravenous fluids in the emergency department especially for pediatric dosage adjustment for epilepsy. Nanomicellelar delivery system might be the possible solution. The objective of this work is to formulate diazepam loaded nanomicelles for the intravenous route. The nanomicelle formulation based on micellar formation in dry form. The influence of types and concentrations of incorporated surfactants and co-surfactants were investigated. The results thus obtained showed that concentrations of surfactant and co-surfactants had a apparent impact on the globule diameter, PDI, , entrapment efficiency, percentage of drug loading, diazepam release profiles and viscosity of the produced nanomicelles formulas. Subsequently parameters play a crucial part in optimization diazepam nanomicelle formulas were studied. And optimized nanomicelles showed a mean globule diameter of 69.38 nm with a PDI of 0.135 on Zetasizer nanoZS TM and a spherical shape on Supra-55VP TM TEM imaging as well as exhibited the best release profile. Three-month stability evaluation of globule size showed stable formulation, which implied that nanomicelles was quite stable when kept at room temperature. Stable diazepam nanomicelle has been attained.
