This study aimed to use molecular methods on clinical oral infection samples to detect C. glabrata virulence gene, hyphal wall protein 1, HWP1, a periodontitis is an inflammatory condition which gradually destroys the supporting structures of teeth. A major concern in dental health, it has a high prevalence rate and significantly affects life quality. Oral candidiasis lesions often develop as a complex biofilm containing C.glabrata. Materials and Methods: Eighty oral swabs were collected from periodontitis patients attending a dental health center and outpatient clinics in Ad Diwaniyah City, Iraq the study was beginning from April 2023 to September 2024. The ages of patients ranged between eighteenth to forty nine years old provided samples taken with disposable cotton swabs. Samples were then cultured on various media like CHROMagar and Sabouraud’s dextrose agar for isolation purposes of Candida glabrata. The results found Among the 80 samples, 18 (22.5 %) were identified as Candida glabrata with the HWP1 gene detected in PCR assay. The Conclusion found the detection of the HWP1 virulence gene in most C. glabrata isolates indicates that the HWP1 protein may play a significant role in the infection's pathogenesis
